This difference in energy indicates complex stability where a negative normalized energy (ΔG
Furthermore, to test the impact of a mutation in residue G339, we analyzed the effect of reverse mutagenesis. We used the generated models and in silico tools to test the effect of reverse mutation at residue G339 on complex stability in subvariants BA2.75, XBB, and XBB.1. They have an aspartic acid residue at position 339.

Duchenne muscular dystrophy (DMD) and Becker muscular dystrophy (BMD) are rare neuromuscular diseases caused by a mutation in the gene that produces dystrophin, which is responsible for maintaining muscle properties [1]. The lack of dystrophin leads to the progressive degeneration of muscle fibers, which become connective tissue and fat [2]. The worldwide prevalence is approximately 0.5 per 10,000 male individuals for DMD and 1 per 3500 male individuals for BMD [3].

The third most prominent mutation identified was N501 Y, in which asparagine (N) was replaced by tyrosine (Y) at position 501 of the S protein (Multimedia Appendix 1). The transmissibility of the virus harboring the N501 Y mutation (located at the receptor-binding motif) increased by 70%-80% and this mutation also improved the binding affinity of the virus onto host cells [27].

Quantifying how the landscape of SARS-Co V-2 mutations in the SARS-Co V-2 genome changes over time can reveal how viral evolution may be associated with specific patterns of mutation burden, mutation types, and genomic locations of mutations, as well as different selection pressures.

A real-time server is set to build and send sequence or mutation files to the Stanford Database and return the expected analysis outcome. Different variables are captured immediately into an Excel sheet (Microsoft) and available for statistical analysis of the app.
Application system design and preliminary testing procedure: Model indicating all probable activities by the user of ARVPredictor and predictable respective responses by both real-time server and Stanford HIVdb.

Response: Information regarding the Romanian patient bearing the ORF8 mutation was added in the Results section.
3. From a public health standpoint, how did the authors deal with the “news” of the new variant? Was there any communication with local officials or support for contact tracing?
Response: This information was added to the text. In addition, we mentioned that our laboratory is 1 of 4 (at the moment of writing the paper) that reports weekly genomic data to government agencies.

Emergence of the First Strains of SARS-CoV-2 Lineage B.1.1.7 in Romania: Genomic Analysis
One such mutation is present only in the sample originating from Suceava, affecting the ORF8 protein, where a stop codon is gained by changing a C nucleotide to a T nucleotide in position 27,945 in the genome. According to Co V-Glue, this mutation has already been encountered in over 580 samples from April to October 2020. Of these, 73% (n=313) belong to specimens collected in the United Kingdom [3].
